The use of gold in medicine, primarily in cancer diagnosis and therapy, has led to other biological applications, in the form of gold nanoparticles (AuNPs), to act as drug delivery agents, as contrast agents in computed tomography, in the production of bioimages, and as an aid in gene therapy, due to their size and properties. It is believed that biomacromolecules from organisms interact with the core of this metal through different mechanisms, resulting in biological protection. However, gold nanoparticles share the same concerns as other metallic nanoparticles regarding toxicity, which deserves to be investigated. This study aimed to evaluate the mutagenicity index (MI ≥2) of AuNPs produced with curcumin (Cur-AuNPs) using the Salmonella/microsome test, the first recommended and validated test for toxicity assessment. Preliminary toxicity test of Cur-AuNPs was carried out using TA98 and TA100 tester strains, and mutagenicity test using TA97a, TA98, TA100, and TA102 tester strains, with and without metabolic activation (S9), in triplicate, using the pre-incubation method. Positive, negative, and spontaneous controls were used to monitor the quality of the test. Concentrations of Cur-AuNPs (µg/plate) of 1.25, 2.5, 5.0, 7.5, and 10 resuted as negative for mutagenicity (MI < 2.0) in all four studied strains. The absence of mutagenicity below 10 µg/plate allow us to point out a safe range for the Cur-AuNPs according to the mutagenicity parameter.
