Equine reproduction plays a strategic role in genetic improvement and economic sustainability of the equestrian sector, making it essential to develop tools capable of more accurately predicting the fertility potential of stallions. In this context, seminal plasma has attracted growing scientific interest for containing molecules directly related to sperm physiology and reproductive performance. The present study aimed to review the literature on seminal plasma proteins in stallions, with emphasis on HSP-1 and CRISP-3 proteins, highlighting their biological functions and possible application in the evaluation of stallion fertility. To this end, a bibliographic review was conducted covering aspects of equine reproduction, composition and functions of seminal plasma, sperm maturation, and contributions of omics sciences in the identification of molecular fertility markers. The literature demonstrates that seminal plasma does not act solely as a transport vehicle for spermatozoa, but also as a biologically active medium capable of modulating sperm motility, viability, and capacitation. Proteomic studies indicate that alterations in these proteins may be associated with differences in fertility potential and semen freezability, reinforcing their value as possible biomarkers. It is concluded that seminal plasma proteins represent promising tools for improving the andrological evaluation of stallions, contributing to more accurate selection of breeding animals and to the development of strategies that optimize reproductive rates in the equine species.
