Resumo

This study evaluated the influence of storage time and peptone water addition on the viability of microorganisms representative of the fecal microbiota of nursery pigs. Individual fecal samples were collected directly from the rectal ampulla of six piglets and subjected to two storage conditions  (with and without the addition of 0.1% peptone water) and analyzed at 0, 6, 12, 24 and 36 hours. Counts of Lactobacillus spp., Bifidobacterium spp., and Escherichia coli were expressed as log₁₀ CFU/g. A significant effect of storage time was observed for Lactobacillus spp. counts (P < 0.01) and Escherichia coli counts (P < 0.01), whereas Bifidobacterium spp. abundance did not significantly change (P > 0.05). For Lactobacillus spp., there was a reduction in counts up to 24 hours, followed by an increase at 36 hours. In contrast, the abundance of Escherichia coli initially increased during the first 6 hours but subsequently stabilized. The addition of peptone water did not significantly influence the counts of any of the microorganisms evaluated (P > 0.05), and there was no significant interaction between the factors. Storage time is a critical factor in microbiological analyses, especially for Escherichia coli, and immediate processing of the samples or their conservation under refrigeration is recommended.
