The mutation pattern of the MDR-1 gene in dogs has a strong breed-specific character, especially in Collies, Australian Shepherds, and Shetland Sheepdogs, and less commonly in Border Collies, German Shepherds, and Old English Sheepdogs. In Brazil, there is still limited data regarding this mutation. This study assessed the incidence of the MDR-1 (ABCB-1) gene mutation using PCR in 236 dogs from nine different breeds. Most animals had normal results (87.1%), while 13.5% were heterozygous and 0.9% were homozygous for the mutation. According to breed, heterozygosity for the mutation was identified in 54.5% of Collies, 35.3% of Australian Shepherds, 40% of Ovelheiro Gaúcho, 25% of Swiss Shepherds, 13.9% of Shetland Sheepdogs, and 5.4% of Border Collies. One Collie and one Australian Shepherd were homozygous. This study concludes that the MDR-1 gene mutation is present in Brazilian dogs, highlighting the importance of PCR screening in predisposed breeds, particularly before the administration of chemotherapeutic agents and drugs that act on P-glycoprotein, in order to prevent severe pharmacological toxicities.
