Abstract

Introduction: Sirtuins are essential protein deacetylases that influence aging and cellular health, with varied functions depending on cellular location. Among sirtuins, SIRT1 stands out for its role in various health and disease conditions. Objective: This study aims to highlight the impact of sleep, diet and exercise on SIRT1 activity, provides specific insights into possible interventions to prevent or reverse health conditions related to aging. Methods: A literature review was carried out in the PubMed and Web of Science databases, using descriptors from the Medical Subject Headings (MESH) of the National Library of Medicine. The search included review articles and clinical trials, without restrictions on the publication period, focusing on the keywords: sirtuins, sirtuins 1, sirtuins 1 and sleep, sirtuins 1 and diet, sirtuins 1 and physical exercise. The review was carried out between January and December 2024. Final Considerations: Sleep quality, a balanced diet and physical exercise are crucial determinants for the modulation of SIRT1. Sleep disturbances decrease SIRT1 function, while instructions such as CPAP use and melatonin administration can restore its activity. Diets rich in AGEs inhibit SIRT1, but antioxidants can reverse these effects. Physical exercise improved the expression of sirtuins, promoting mitochondrial health and resistance to oxidative stress. We conclude that modulating SIRT1 through lifestyle disciplines is fundamental for more personalized and effective medicine, promoting health and longevity.
