Abstract

Species of the genus Monteverdia, among them, M. quadrangulata, have been traditionally used in Brazil to treat infections and inflammation. The objective of this study was to investigate the effect of polar extracts of Monteverdia quadrangulata on viral particles and the replication cycle of Zika virus (ZIKV). The anti-ZIKV effect of ethyl acetate extracts from leaves (LAE) and branches (TAE) of M. quadrangulata was investigated in vitro, and the median effective concentration (EC50), selectivity index (SI) and mechanisms of action (virucidal activity and antiviral activity in the adsorption, internalization and post-infection stages) were evaluated. The EC50 values of the LAE and TAE extracts were ≤ 50µg/mL. LAE was the most selective (EC50 = 29 ± 9µg/mL, SI = 31) with significant virucidal activity. LAE also reduced viral load by 4 log units at different times after infection. Probable catechins and epicatechins, previously identified as constituents of LAE, act synergistically at different stages of the viral cycle, inactivating extracellular ZIKV and interfering in the viral replication cycle.
