Abstract

Trigonelline (T) is an alkaloid present in the leaves of Bauhinia sp., popularly known as "cow's hoof," used as an infusion for diabetes. Blood samples from diabetic patients have shown alterations in erythrocyte aggregation parameters and in the mechanical properties of red blood cells (RBCs). The objective of this study was to evaluate the hemorheological action of (T) in an in vitro hyperglycemia model. Therefore, RBCs obtained from healthy donors were washed and glycated at different glucose concentrations, and then incubated with T solution in phosphate buffered solution at 0.25 and 1.0 mg/mL. An inverted optical microscope attached to a digital camera was used to observe the samples. The aggregate shape parameter (ASP) and the isolated cell coefficient (CCA) were obtained through digital image analysis. The erythrocyte aggregation phenomenon was evaluated using an optical chip aggregometer. The images obtained show large, globular erythrocyte aggregates in the glycated samples, which reverse with the addition of T, and the CCA decreases significantly with T at 1.0 mg/mL. The aggregation half-time and aggregation amplitude decrease with the addition of T for both concentrations. These results provide information on T's hemocompatibility and potential activity on the red cell membrane, making it useful for use as an adjuvant in the treatment of diabetes or as a preservative for concentrated RBCs for transfusion.
